If you're interested you should. Considering your penchance for roleplaying I think you would do well in writing fan fiction for the movie that utilizes your own unique twists.

I've got it. I know what really should have been a part of the Mario movie. The boos! Mario and Luigi could have discovered several secret passageways in Koopa Tower and the boos could have been lurking in there.

Perhaps Blarg's initial appearance could just be eye's and the top of his head poking up out of the lava briefly
Or maybe used as a weapon. Magi invents a "Techno seed" that when fired into lava forms Blarg. Or some mystic talisman falls into some lava and makes a Lava Golem. Similar approaches could be used with Ice to create Ice Golems, based on the Slipice enemies from Mario Brothers
Fighter Flies Basically mutant insects that look like a cross between flies and grasshoppers. Crabs could be seen with them, maybe vaguely bipedal like Mirelurks. But I couldnt really see much purpose for them other than a background joke.
For the Mole guy you could get some Little People to play them. Instead of the thief angle perhaps a Full Metal Jacket-esque "Tunnel Rat" based on the fighters of the Vietnam War. Perhaps over the top soldier caricatures like Sgt Rock and Nick Fury
Bullet BillCould be an ICBM, missiles on an aircraft, a visually guided smart missile or even a aerial drone.
R.O.B. Robotic Overseer Construct. A robotic assistant created by either Doctor Magi or Proffessor Gadd
Virtual Boy could be used as a viewer, similar to the one seen on Spock's station on Star Trek. Someone uses it then looks up rubbing their eyes saying "Damn I hate using this thing."
Light Zappers could also be used as standard weapons.
Game Boy can be used as portable scanners. Might even have a face on it and talk to characters ala the Captain N character. Cartidges serve as programming enchancements. For instance for a medical scanner, Dr Mario is used.
If Warp Pipes are used as interdimensional portal, perhaps one could malfunction and bring in minor characters from other games like a Met from Megaman, a Metroid, a Tektite and a duck from Duck Hunt. Or be shown brief glimpses into other video game wordls. The Tektite tries to jump on Peach, and Mario kicks it away. Peach tells him "I could have handled that myself, you know:" To which Mario quips "Well Excuuuuse ME, Princess"
Peach doesn't have to be Daisy's sister. They could be cousins, and Peach was there visiting, and her family left before Koopa staged his coup
